Just pulled a 4cyl, extremely low mile Mercedes 4cyl and 7 speed auto and full harness from a totaled Sprinter Van. Got the noggin turning. Anybody have a good reason not to go down this rabbit hole?

Not a lot, but the 7 speed makes it seem like it does. In those godawful vans, it'll put you back into the seat a bit, AND get 25+mpg.

Also, that harness is the entire vehicle harness unfortunately. Benz, in their infinite wisdom, did not deem it necessary to separate the chassis and engine harnesses...
 
That harness...

Just go LS3 crate engine.

If you're blowing smoke with a diesel, you may as well be chopping up 100$ bills and snorting them through 100$ bills. A well tuned diesel engine will always be preferred, at least until electrics are up to speed. The Diesel cycle is just inherently more efficient, unfortunately there are a bunch of damn kids and old Chrysler engineers who have given them a bad name.

Short answer, no.

DEF has really nothing to do with the power it puts out.
The DPF on the other hand yes. But this is a rather small engine and you can't expect miracles out of it.
20-30 more HP at the most and that's pushing it.
Torque around 300.

Running without and emission restrictions and a computer tune should get you there.
185-190 HP and roughly 300-320 TQ all on stock internals.
