Trying out new spare key in my 1983 FJ40. Passenger door unlocks smoothly with keys. Driver's door does not unlock with either key. Key doesn't even turn while in slot.

Driver's door manual lock make a "creaking springy" sound.
Before I take the 3 screws out of the driver's door and go digging are there any dos and don'ts or quick tips & fixes?
Thank you.

It's not clear what's happening here. You have two "spare" keys that work in the ignition and passenger door but not in the driver's door, and primary key works in all doors and the ignition?
Or, you have NO key that works in the driver's door lock?
If it's the second scenario, I'd get new lock cylinders for both doors and have one key for the doors and one key for the ignition. Or, the simple path would be to take Pighead's advice.

I have no key that works in the driver‘s door.

I don't know about availability of lock cylinders for FJ40 to purchase (edit, see below), but do have experience switching them out in a 4Runner and a Nissan Hardbody from the 90's. It wasn't difficult for those vehicles so my guess is that it would just as easy if not easier with an FJ40. Hopefully someone else will chime in. Did a search and most are taking the doors off because there’s a tiny spring clip that is easily dropped and lost into the bottom. A few with small hands did it without door removal.

Typically you need to remove the interior door panel, but not the entire door.

Rekeying a lock isn't all that hard.

I never lock my FJ40 - anyone can open it in like 5 seconds with a coat hanger threw the door or the kick vent.

When asking for help year model market helps get better answer. Look at the door schematics at like SOR.com

I have not locked mine yet. Always have wallet, phone and sunnies with when away.
Thanks for the reminder. Editing original post now.
