What is the best tool for mapping routes w/o using highways. I’m driving from Indy to Florida and want to take a routes through the countryside.

Goggle has an option to avoid highways.

In my 200, I use Google maps and used the avoid highways feature on a camping trip west 2 years ago. It worked well, but I occasionally overroad its decisions.
 
Not sure what you're looking to do but I have some suggested roads if the idea is to wander a bit to keep it scenic. If you're trying to make time then the Google avoid highways suggestion is about as good idea as any.

If you're willing to first travel SE and cut down towards Northern KY via US 52, KY 10 to KY 165 are nice scenic roads that run atop the hillsides of the OH River Valley once you get SE of the Cincy metro and get to about Mt Olivet, KY. This then allows you to meander down to the Winchester/Mt Sterling area however you choose. At that point just stay east of I-75 and pick a N/S road and you can't really go wrong. Roll south into the vicinity of Knoxville and then B line towards Deals Gap via US 129. Eventually find US 19 and hit up the Blood Mountain area in northern GA. Keep tracking to the ATL metro and then pick your poison.

Daylight obviously isn't your friend this time of year and I don't know how long you're trying to take. That eastern TN/western slice of NC into northern GA is primo though.
 
I use an app called Roadtrippers, let's you looks for all kinds of things, parks, sights, photo ops, almost anything. Ill look for the neat stuff I can find until my destination. Then just end up all over. That's what I've done heading to LCDC and see so much cool s***.
