I was driving on the Highway the other day goin uphill and the car wouldnt engage the 5th gear, after getting above 3000 rpms it made a grinding noise, like it tried to engage it but didnt work, anyone has a clue of what could it be? Its a sealed unit so I can't inspect

It isn't sealed and you can inspect. FAQ has your back. Check fluid level and condition before anything.
Good luck, scary I know, but it's a strong trans and shouldn't be a major issue..

How should I inspect it? Removing the pan underneath?

Thanks for replying!

Your transmission has 2 plugs on the pan, one to drain and one to set level at a certain temperature (about 100F) and while engine running. Also a 24mm plug on the side to fill up. WS Toyota fluid only, new washers are recommended, and the procedure is in the FAQ. Welcomed to search for it as well, discussed several times in detail. @2001LC has detailed info on several of his threads, great to follow him.
Also search for the available PDF service manual somewhere here..
You might have to lower the pan if there's a stuck solenoid, that's after a detailed diagnosis, of course.

Just searched FAQ and couldn't find a direct access to the transmission fluid check procedure for the 5 speed @Trunk Monkey, maybe I missed it?

That's as good as FAQ for my use.

Yeah Ive been trying to find it and only found for the transfer case so far, But I really appreciate your replies!

I guess I'll have to write one then.. I'll go polish my tools and get to it.
One more thing that I forgot to mention. After pulling over and doing a quick inspection I drove back home going 50 mph or below. My question is, would it be safe to drive it after checking it just to try it out? since apparently all the other gears were engaging.

I'd do a drain and refill first, that will get about 3-4qts of fluid out and assure the level is correct.
Then take it out for a run to see if that fixed the issue. If yes, schedule another drain and refill soon, and keep it as a maintenance schedule every 3-4 engine oil changes.
If not, and issue persist, get it scanned for codes. A regular $20 scanner might not read these, you might need a pro level scanner, maybe a friend with one? Not sure if Techstream is available to you. But a $125 diagnosis by a pro scanner beats a $4k trans rebuild for an issue that might be sorted with some elbow grease and a sensor/solenoid swap..

edited to add, did you check that the 2nd/pWR buttons are off?

Yeah I have a ultra gauge but just reads the transmission fluid temperature, Ive been asking for one that can read that but it might be a good investment. Both buttons were off

What ATF #1 temp have you been seeing in what OAT/road/driving condition?

The 04-07 were underfilled at factory. I'd do a full 12 qt flush, ASAP. Set level with AT #1 temp at 96f -115F. I like to set, closer to the lower temp.

Since 2006 your AT, possibly had a drain and fill. Most times, tech take out 3 qts add 3 qts. Or set level to old FSM temp or dash light method. Which those were used, before Toyota TSB lowered the temp. So level likely still low. Additionally, no way to know, what fluid was use. Very often shops use cheap bulk. So you end up with low level and a mix of ATF in AT.

I use 12 qts: Mobil MV full synthetic ATF (rated for Toyota: II, III, IV and WS ATF), which can be hard to find locally. Or I use Toyota WS ATF.
