Don't close the vents...not only do they vent but they also provide depressurization for the cabin.

I haven't opened these up, but if they are like most vehicles, they are one-way - out. They don't help bring in fresh air unless there is a two-way door as part of the A/C system.
I bigger concern with sealing them could be CO.
Now, the methane problem you referenced above is the most likely and more common scenario we all face, especially with teenage boys
Correct, they're flapper valves. The air flows front to rear. Blow air in through the HVAC and it goes out through the vents. If they're some how blocked or closed then there wouldn't be much flow happening.
