In the very near future I will be installing a water storage tank directly behind the front seats. Overall, full, the weight will approach 150lbs. I would like to strap it down for safety reasons and it appears that new holes will have to be drilled into the floor and nuts tacked to the under side of the floor.

Has anyone done this? Any advice on what to look out for? The obvious is to not drill into a fuel line, brake line, etc. Did you reinforce the area around the nuts for strength?

I have not read about this being done on any of the forums I read, so sny info would be appreciated.

Honestly, with a tank behind the front seats you are loosing use of the rear seats for the most part. Why don't you take out the rear seats and use the existing seat and or seat belt locations to secure your tank. Get a little creative and add storage of some kind.
If I may, what's the purpose of said water tank any way?

I guess I better make myself more clear and I don't mean that sarcastically. I haven't had my rear seats in in over a year. Long gone for storage/sleeping platform that runs to just behind the seats. Though now we have a RTT and don't use it for sleeping anymore.
So after a couple of years of camping, the wife and I sat down and figured out how to cut down on the many stuff sacks, duffels, water jugs, etc. that take up so much room. We also have two dogs that travel with us.
Part of the consolidation is to build a water storage tank that fits as described above - 17 gallons. We will have a spigot at the back door that we will get our water from - pressurized by a Shurflo 12v pump. I will also have a quick disconnect point where I can connect a shower hose and have showers on the trail.
To get some kind perspective here is a pic of the part of the rear where the seats should be. I have the side panel off to relocate my ham radio and the 12v pump in the space above the wheel.

Anyway you can see the tank, in it's early stages, where it will sit. It's the white thing on the right. I have 4" high tool storage at the back, with our Weaco fridge on top at the side and in front is where we have our hidden storage area/dog platform.
