Recently bought a norcold fridge. Now I see that my rear power source is only hot when the key is on. This is in my wife's Honda pilot. Should I run a fused always hot line from the battery to the cargo area ,or will the items in the fridge stay cold over night?

Run power back there. My wife has the equivelent Honda and the power back there is terrible. I ran an inverter running a laptop and got the power socket overheated and blew the fuse.

Run a 12g wire back there.

I don't have experience with a norcold fridge, i have had experience with a truckfridge in my vw camper. on a decent auxiliary car battery, it would keep it running overnight into the morning (6pm- 9am) most of the time. I had to run a huge deep cell battery to keep the fridge going for longer than overnight.

I'm not sure that the power draw is for your Norcold, but if it is anything like my Engel it isn't much. The power in my Tundra is timed - stays on for probably a 1/2 hour. I ended up making a cigarette lighter connection that clamps directly to the battery. Runs all night with no issues. I recommend getting an insulated cover to help cut down on cycle times as well.

I found a manual and it says to use 10 gauge wire. So I am going to do like you guys suggest and run a fused wire to the cargo area. Going on a 2 week trip It will be so nice not to deal with ice. Going to Spencer's graduation from basic training at Ft. Jackson SC. Then down to Florida for a few days, Then New Oleans for 2 days
