So I dropped my spare tire and noticed some rubbing along the sidewalls with some material loss. Is it possible to crank the tire up too much and for this to happen? I’m obviously worried about the integrity of the tire now.

I don't see an issue with that tire sidewall
MHO would be it was loose not tight, than the tire would be rubbing causing wear, up tight the tire wouldn't move

Hard to tell from the pictures if that's just a scuff or the rubber has worn. I don't have a spare tire in the stock location anymore but I recall the cross bar had some kind of foam to protect from that. ( or may be I am thinking of another vehicle like my GX) Either way if you're concerned you could put something up there to help protect from that ...And then just apply normal force when securing the tire don't crank down on it but make sure it doesn't wobble around when you shake test it
