I have a 74 FJ40 and reinstalling the charcoal canister to deal with gas fumes. My question is, can I run the hose line from the charcoal canister to this plugged port on the side of the carb ? This is not the oem carb since the FJ40 has been de-smogged. Hoping to vacum the fumes into the carb post filter.

Not sure on specifics as I'm more familiar with the Aisan carb, but I believe you would use that port to actuate a vacuum switching valve which would allow a vacuum to be pulled from your intake manifold to the charcoal canister outlet.
