yes, drill it out, thread the hole and screw in a threaded plug with sealant applied to the threads. you'll never need to drain it from there, but if that plug falls out, then you'll lose all oil before you know better and ruin the motor. most guys who read about this do it while still on the engine with proper care taken not to contaminate the galley with chips- some guys with engine still in truck. no excuse to not do it with everything apart. they have been known to pop out post rebuild- something about the hot tank and type of material of the plug. couldn't cost much to have done- it's about free to do it yourself.
I hope that block gets seriously cleaned before assembly - clean engine is good engine . Paint looks good , btw.

I haven't read through the whole thread recently, but if you haven't already, get the rotating assembly balanced while you are at it. I'm just getting my 2FE running right now, and holy crap the difference. Spins 4,600 like it's nothing!
Here's a good article explaining the process. Basically, you are making sure things that spin (crank, rod, piston) all match in weight and the crank is balanced along it's length. If you have already started assembly, it may be too late unless you want to backtrack. It cost me about $200 extra IIRC, at the machine shop, and I think it's the best money I've ever spent on my 40.

Also, MAKE SURE you have the right freeze plug (i.e. toyota sourced only) in the back of the block behind the cam. One that is too deep won't allow the cam to sit far enough in the block and will kill lifters in a hurry as well as the cam itself.
