In-fact you should not be able to blow air (very low pressure) from the carb side to the tank. If you can that means the check valves are stuck open. If they are stuck open (even one) than the fuel pump will not be able to create a vacuum thus not be able to draw fuel from the tank.
You could also try pouring fuel into the supply line and filling up the filter first. But I still think the check valves are stuck. Try blowing air through the line before buying any new parts.
I have seen situations where the check valves (there are two) have frozen due to varnish. To resolve this issue I have blown air (at low pressure) from the supply side of the pump forcing the valves to unstick.
Other issues could be you are not waiting long enough for the pump to prime itself...
All of my brake/clutch cylinders that I bought from CCOT died within 1 year. After that I only order OEM brake/clutch master/cylinders.

You cannot tell just by looking at the piston, remember that the piston rises to TDC twice. That also means that the flywheel will say TDC twice. I would flip the rotor 180 degrees and try again.

Well, you probably overloaded the internal circuits of the voltmeter. Most voltmeters can't handle that high of a voltage that is being produced at the spark plug.

Thats what I was thinking. Something about if you are unlocked one wheel will keep you from sliding sideways while the other wheel will drive you. But if you are locked the back end will try to slide sideways.

Don't throw your stock carb away, a propery built Aisin(stock) carb will beat any weber/holley. I don't think that its your carb or anything. I would think that for some reason when the fuel tank is full a vacuum is being created inside the tank. I would fill it up and drive it about a mile...
