Engine is a 1973 1F. What's this hole for?

That's for a thru bolt that attaches to the stock exhaust manifold. It's fine as is. Does not connect to inside of intake.
 
Dipstick tube. Your dipstick is missing?

Shouldn't I plug it so it doesn't affect intake vacuum?
That hole does not communicate with the interior of the intake manifold, it has no affect on vacuum.
Put a bolt thru it if you don't like looking at an empty hole.
